Madame Web 2024 follows the tale of a young woman who becomes the savior of three teenage girls. In 1973, Constance Webb ventures to the Peruvian Amazon while pregnant to study healing spiders. Despite challenges, she finds the spider, only for her guard, Ezekiel Sims, to take it for its powers. Tragically, Ezekiel shoots Constance before the Las Ara?as tribe comes to her aid. She gives birth to a baby girl before passing. Three decades later, Cassie Webb, Constance's daughter, discovers her future-seeing ability while working as a paramedic in NYC.
