Before fumarylacetoacetate hydrolase (FAH), nitisinone inhibits 4-hydroxyphenyl-pyruvate dioxygenase, a tyrosine catabolic enzyme. The inhibition of tyrosine metabolism by nitisinone lowers the accumulation of maleylacetoacetate and fumarylacetoacetate in HT-1 patients.
