Nitisinone: Dosage

The recommended dosage is between 1 and 2 mg/kg, administered twice daily. First-time users receive a dose of 1 mg/kg/day in the morning and the evening. The nitisinone drug should be started by a medical professional with competence in treating HT-1 patients. The nitisinone dosage can be altered for every patient. If after one month after starting treatment, erythrocyte PBG-synthase activity, urine 5-ALA, and urine succinylacetone do not return to normal, the dose of nitisinone may be increased to 1.5 mg/kg/day. If erythrocyte PBG-synthase activity, urine 5-ALA, and succinylacetone remain elevated and plasma succinylacetone has not normalized after three months in patients taking 1.5 mg/kg/day, the dose may be raised to a maximum of 2 mg/kg/day.
